PPAR-a Activation Attenuates Angiotensin II-Induced Vascular Inflammation, Arterial LDL Accumulation and Endothelial Layer Permeability in Mice

چکیده
The peroxisome proliferator-activated receptors (PPARs) have been shown to be important in modulating vascular inflammation and atherosclerosis. Angiotensin II (Ang II) is known to promote vascular inflammation and accelerate atherosclerosis. We demonstrated that these vascular changes induced by Ang II are associated with up-regulation of pro-inflammatory genes, as well as down-regulation of PPAR-a expression. We hypothesized that a PPAR-a agonist would have vasculoprotective effects in a mouse model of Ang II-induced atherosclerosis by: 1) attenuation of pro-inflammatory mediaTham-vol5no1 3/19/05 10:59 AM Page 1

Endothelial Vasodilator Angiotensin Receptors are Changing in Mice with Ageing
Background: The vascular function of Angiotensin II-type-2 receptors in adults is controversial. We sought their location and function in mouse aortic rings at young and old mice. Materials and Methods: Male C57Bl mice (aged 4 and 14 months) were killed by CO2. The descending thoracic aorta was cleaned and dissected into rings. Aortic rings were mounted in Krebs’ solution at 37 °C an...
متن کاملThe Higher Response of Vascular Endothelial Growth Factor and Angiotensin-II to Human Chorionic Gonadotropin in Women with Polycystic Ovary Syndrome
Background This research investigated the response of vascular active factors, vascular endothelial growth factor (VEGF) and angiotensin-II (AT-II) to ovarian stimulation during 24 hours in patients with polycystic ovary syndrome (PCOS). MaterialsAndMethods In this clinical trial study, 52 patients with PCOS and 8 control cases were stimulated with human chorionic gonadotropin (HCG) on the 4th ...
متن کاملZoledronate attenuates angiotensin II-induced abdominal aortic aneurysm through inactivation of Rho/ROCK-dependent JNK and NF-κB pathway.
AIMS Abdominal aortic aneurysm (AAA) is a life-threatening disease affecting almost 10% of the population over the age of 65. Nitrogen-containing bisphosphonates (N-BPs) have been shown to exert anti-atherogenic and anti-angiogenic effects, but the potential effects of N-BPs on AAA remain unclear. Here, we tested whether a potent N-BP, zoledronate, can attenuate the formation of Angiotensin II ...
متن کاملEnhanced immune system activation and arterial inflammation accelerates atherosclerosis in lupus-prone mice.
OBJECTIVE Premature atherosclerosis is a characteristic feature of systemic lupus erythematosus, a prototypic autoimmune disease. The principle cellular and molecular mechanisms which underlie such accelerated atherosclerosis are indeterminate. METHODS AND RESULTS The pathophysiology of lupus-mediated atherogenesis was evaluated in a novel animal model involving transplantation of bone marrow...
متن کاملEffect of Angiotensin II on Blood Flow in Acute and Chronically Inflamed Knee Joints of Rabbits: The Role of Nitric Oxide
Background: Angiotensin converting enzyme (ACE) upregulation in stromal cells of joints affected by rheumatoid arthritis may lead to higher tissue angiotensin II that is a vasoconstrictor and mitogen factor. To date, the role of angiotensin II on regulating blood flow in inflamed joints has not been studied. Methods: Acute and chronic joint inflammation was induced in rabbits by intra-articular...
